Effects Of Topical Vitamin-e And Ascorbic Acid On Keratocyte Apoptosis After Mechanical Epithelial Debridement In Rabbit Eyes

Material and Methods: Twenty New Zealand white rabbits were used divided into 4 groups and only one eye of each rabbit was deepithelialized mechanically within a 7mm zone by using a blunt (Kimura) spatula. Group 1 was not treated with any of the vitamins and acted as a control group. One drop of vitamin E (1IU/mg, 50IU/drop) was applied to the cornea 2-3 minutes before epithelial removal in group 2. Group 3 received topical vitamin-C (10% (w/v) ascorbic acid) just after epithelial removal and lastly group 4 was treated with topical vitamin-E before and vitamin-C after the epithelial debridement. Central corneal tissues were removed and TUNEL assay was used for the demonstration of the apoptotic keratocytes 4 hour after corneal surgery. PMN leukocyte densities were also assessed. ANOVA test was used for the statistical evaluation of apoptotic cells and PMN cells between all groups.
Results: Control group had the highest number of apoptotic keratocyte (ANOVA: p<0.05, Post Hoc test: p<0.05 for all three groups). On the other hand, there was no significant difference between each vitamin group (Post Hoc test: p>0.05). There was no statistically significant difference between groups in terms of PMN cell number (ANOVA: p>0.05).
Conclusion: Mechanical epithelial debridement induces keratocyte apoptosis which can be prevented by vitamins E and C at least partially.
